A search for RFLP markers to identify genes for aluminum tolerance in maize Braz. J. Genet.

The objective of this study was to identify restriction fragment length polymorphism (RFLP) markers linked to QTLs that control aluminum (Al) tolerance in maize. The strategy used was bulked segregant analysis (BSA) and the genetic material utilized was an F2 population derived from a cross between the Al-susceptible inbred line L53 and Al-tolerant inbred line L1327. Both lines were developed at the National Maize and Sorghum Research Center - CNPMS/EMBRAPA. The F2 population of 1554 individuals was evaluated in a nutrient solution containing a toxic concentration of Al and relative seminal root length (RSRL) was used as a phenotypic measure of tolerance. ANALYSES OF GENETIC VARIABILITY IN LENTINULA EDODES THROUGH MYCELIA RESPONSES TO DIFFERENT ABIOTIC CONDITIONS AND RAPD MOLECULAR MARKERS BJM

The growth of thirty-four Lentinula edodes strains submitted to different mycelial cultivation conditions (pH and temperature) was evaluated and strain variability was assessed by RAPD molecular markers. The growth at three pH values (5, 6 and 7) and four different temperatures (16, 25, 28 and 37ºC) was measured using the in vitro mycelial development rate and water retention as parameters. Mycelial cultivation was successful at all pH tested, while the ideal temperature for mycelial cultivation ranged between 25 and 28ºC. The water content was lower in strains grown at 37ºC. Characterization of rhizobia that nodulate Arachis pintoi by RAPD analysis BJM

The genetic relationships of 85 Arachis pintoi nodulating Rhizobium strains were determined using the random amplified polymorphic DNA (RAPD) methods. The analysis included 75 strains isolated from Cerrado soils and 10 other ones of different origins. The results indicated that there is a high level of similarity between these strains and that geographic distribution may affect their phylogenetic relationship. In addition, the results allowed the selection of the most suitable primers for characterisation of these Rhizobium strains which will be useful for implementation of competitiveness studies in Cerrado soils.

Effect of ABA and GA3 on protein mobilization in embryos and cotyledons of angico [Anadenanthera peregrina (L.) speg] seeds during germination BABT

In this work, a woody species [A. peregrina (L.) Speg.] was studied in order to observe the effect of ABA and GA3 at the biochemical level during the process of seed germination. Embryos incubated in sucrose solution containing ABA and/or GA3 were analyzed through SDS-PAGE to observe the mobilization pattern of storage proteins during the beginning of germination. Effect of Baculovirus spodoptera isolates in Spodoptera frugiperda (J.E. Smith) (Lepidoptera: Noctuidae) larvae and their characterization by RAPD Neotropical Entomology

The total of 22 Baculovirus isolates surveyed in different corn producing regions in Brazil were used against fall armyworm, Spodoptera frugiperda (J.E. Smith). The viruses were purified and their suspensions were used to feed fall armyworm larvae from 4th and 5th instar. The mortality rate was checked daily and the infected larvae were frost after death, what generally occurred between the 5th and 7th day after virus ingestion. High temperatures can affect the survival, establishment and symbiotic properties of Rhizobium strains. Bean nodulating Rhizobium strains are considered particularly sensitive because on this strains genetic recombinations and/or deletions occur frequently, thus compromising the use of these bacteria as inoculants. Eficiência de índices fenotípicos de comprimento de raiz seminal na avaliação de plantas individuais de milho quanto à tolerância ao alumínio PAB

O objetivo deste trabalho foi testar a eficiência do comprimento relativo de raiz seminal (CRRS) e do comprimento líquido de raiz seminal (CLRS) como indicadores fenotípicos quanto à tolerância ao alumínio na avaliação de plantas individuais de milho. Plântulas de genótipos tolerantes e suscetíveis ao Al foram submetidas a solução nutritiva contendo nível tóxico deste elemento, por um período de sete dias, após o qual, determinaram-se os valores de CRRS e CLRS. Fall armyworm, Spodoptera frugiperda (J. E. Smith), is the most important maize insect pest in Brazil and its damage can reduce yield up to 34%. The objective of this work was to utilize the PCR technique to Identify Bacillus thuringiensis (B.t) strains that control S. frugiperda in maize crops. Bioassays showed that 16 strains were very effective against S. frugiperda, 15 of them collected from soil samples and one (T09) obtained at the Institute Pasteur. Indução e análise bioquímica de calos obtidos de segmentos foliares de Coffea arabica L., Cultivar Rubi Ciência e Agrotecnologia

Conduziu-se este trabalho com o objetivo de estudar o efeito de 2,4-D e BAP na indução da formação de calos originados de segmentos foliares de plântulas de Coffea arabica L., cv Rubi, obtidas in vitro, a partir da cultura de embriões, determinar sua curva de crescimento e analisar bioquimicamente, mediante determinação dos teores de proteínas totais, açúcares redutores e padrão proteíco. Segmentos foliares inoculados em meio desprovido de reguladores ou na presença de BAP em condições isoladas não apresentaram formação de calos. Marcadores microssatélite têm sido utilizados na determinação da pureza genética em programas de melhoramento e de produção de sementes, visando garantir a qualidade genética das sementes. Objetivou-se neste trabalho demonstrar, com auxílio de marcadores moleculares microssatélites, que variações na coloração do hilo em sementes de soja, muitas vezes não constituem variação genética. Foram avaliadas sementes de soja da cultivar CD 222 que apresenta sementes com hilo preto, e de duas linhagens (CD 02RV-8444 e CD 01RV-7618), as quais apresentam sementes com hilo marrom. Muitos métodos para determinar a presença de taninos são descritos na literatura, mas nenhum deles é universalmente aceito como ideal ou utilizado de forma unânime. Alguns métodos colorimétricos não diferenciam taninos de outros compostos fenólicos, outros utilizam substâncias que não são adequadas como padrão. In South America, about 66% of the land surface is made up of acid soils where high aluminum saturation is one of the main limiting factors for agriculture. Random amplified polymorphic DNA analysis of effective Rhizobium sp. associated with beans cultivated in brazilian cerrado soils BJM

Efficient bean nodulating Rhizobium strains, isolated from different Brazilian cerrado soils, were characterized by RAPD. This study showed great genetic heterogeneity among R. tropici and R. leguminosarum bv. phaseoli strains and allowed the constitution of genetic clusters, besides indicating the most suitable primers for this characterization. The groups of genetically distinct strains can be used in competitiveness studies to select appropriate Rhizobium strains for bean inoculation in cerrado soils.

The objective of this work was to identify the most competitive and effective Rhizobium strains in order to increase common bean yield by nitrogen fixation as alternative or complementation to the nitrogen fertilization. Variability of isolated colonies in bean nodulating Rhizobium strains before and after exposure to high temperature BJM

Irregular response to bean plants to Rhizobium inoculation has been attributed to among other factors, low competitive ability, low N2 fixation efficiency and genetic instability of the symbiont.
